Robust RNase-Free, Animal-Origin-Free Recombinant DNase I

What is RQ2.0 DNase I?

RQ2.0 DNase I, based on the enzyme from bovine pancreas, is an endonuclease that exhibits activity against both single-stranded and double-stranded DNA. It is essential for removing DNA contamination in in vitro transcription (IVT) RNA production, RNA purification workflows and molecular biology applications.

Why choose recombinant DNase?

Recombinant expression of DNase eliminates animal-origin contamination risks, which when combined with our stringent quality system, ensures the high level of purity and consistency required by diagnostic and therapeutic developers.

How does RQ2.0 DNase I improve RNA production workflows?

The robust formulation and consistent unit-activity definition of RQ2.0 DNase I make it highly reliable for research or regulatory-compliant workflows in RNA manufacturing, sequencing and purification. Consistent lot-to-lot performance provides better digestions and less time optimizing.

Representative data of residual DNA quantified by qPCR of a 250 bp amplicon following digestion with 1 U/μg DNA per the manufacturer's specification in moderate ionic strength, 40mM (Left light blue), high ionic strength, 330mM (Left Dark Blue) or under no magnesium buffer (Right) conditions. Reactions were performed with a model template spiked into a buffer modeled after a typical in vitro transcription reaction conditions including Tris-HCl (pH 7.5), MgCl₂, NaCl, DTT, CaCl₂, spermidine and rNTPs. . R=Roche rDNase AOF/cGMP, N=NEB rDNase, and T= Thermo’s TheraPure rDNase AOF/cGMP. Higher Cq = less residual DNA.

Representative data of residual DNA quantified by qPCR of a 250 bp amplicon following digestion with RQ2.0 rDNase. Reactions were performed with a model template spiked into a buffer modeled after typical in vitro transcription reaction conditions including Tris-HCl (pH 7.5), MgCl₂, NaCl, DTT, CaCl₂, spermidine and rNTPs. Digestion was performed at 37 °C for 5–75 min (Left) or for 30 minutes at room temperature (RT), 37 °C or 42 °C (Right). Higher Cq = less residual DNA.
